چکیده

Procedure – A proprietary formulation of ATCC 33701 Rhodococcus equi known to contain the virulence plasmid was developed and tested on 72 horses. Antibody levels to Rhodococcus equi Virulence associated protein A (VapA) were determined relative to a Standard Plasma. A clinical trial was performed on 28 foals. Seven foals were administered hyperimmune plasma and seven foals were administered purified equine immunoglobulin specific for VapA and VapC one day prior to intrabronchial challenge with Rhodococcus equi strain 103+. Eleven foals were challenged with no prior treatment and three foals received no challenge or treatment. A quantitative anti-VapA antibody enzyme immunoassay (EIA) was developed by using known concentrations of purified antiVapA antibodies to create a standard curve.

Survival of foals with experimentally induced Rhodococcus equi infection given either hyperimmune plasma containing R. equi antibody or normal equine plasma.

The purpose of this study was to determine if colostrum-deprived foals with experimentally induced Rhodococcus equi pneumonia have a decreased severity of the disease and decreased mortality rate when given hyperimmune (HI) R. equi antibody plasma (R. equi titer at least 100 % and virulence-associated protein A [VapA] at least 10000) prophylactically versus when given normal equine plasma (R. e...

Vaccination of Mice with Virulence-Associated Protein G (VapG) Antigen Confers Partial Protection against Rhodococcus equi Infection through Induced Humoral Immunity

Rhodococcus equi is a facultative intracellular bacterium causing severe pyogranulomatous pneumonia, ulcerative enterocolitis, and mesenteric lymphadenopathy in foals aged less than 6 months. Less frequently, this pathogen affects various other species, such as pigs, cattle, cats, and even humans. Although rhodococcosis is treated with a combination of antimicrobial agents, resistance is develo...

Rescue of an intracellular avirulent Rhodococcus equi replication defect by the extracellular addition of virulence-associated protein A

Rhodococcus equi is a facultative intracellular bacterium that can escape from bactericidal mechanisms associated with phagocytosis. Virulence-associated protein A (VapA), encoded on a virulence-associated plasmid, is essential for intracellular survival in macrophages, but its function is not known. Here, we show that the extracellular addition of recombinant glutathione S-transferase (GST)-Va...

Prevalence of the virulence-associated gene of Rhodococcus equi in isolates from infected foals.

The prevalence of the plasmid-encoded virulence-associated gene (vapA) of Rhodococcus equi, as determined by PCR, was found to be 98% in isolates from 154 foals with pneumonia, confirming the strong association of vapA with virulence. The vapA genes from 60 representative isolates were compared by digestion with the restriction endonuclease HinfI, and no evidence of sequence variation was detec...

Evaluation of a multiplex polymerase chain reaction assay for simultaneous detection of Rhodococcus equi and the vapA gene.

OBJECTIVE To evaluate sensitivity and specificity of a multiplex polymerase chain reaction (PCR) assay for simultaneous detection of Rhodococcus equi and differentiation of strains that contain the virulence-associated gene (vapA) from strains that do not. SAMPLE POPULATION 187 isolates of R equi from equine and nonequine tissue and environmental specimens and 27 isolates of bacterial species...
